"spotting solution signal"
we are having trouble eliminating signal from spots that contain only 1X spotting solution (array-it, telechem inc.). we are pretty sure the dyes are not sticking to cross contaminating dna (syber green assay) but cant seem to figure our why its there or how to get rid of it without using super stringent conditions that might shake of real signal. any ideas?

We see exactly the same with the ArrayIT spotting buffer. As we have not come to any other solutions than skipping the ArrayIT buffer I would be very interested if you have come up with any other solution. By the way what is the best spotting buffer for spotting oligos on epoxy slides?

You should use the spotting buffer from MWG biotech. We have compared different spotting buffers and found high background from the ArrayIT spotting buffer. However when we used the spotting buffer from MWG there was absolutely no background. Hope that this will help you.
